Pancreatic cancer organoid is mainly derived from surgical specimens, EUS-guided fine needle biopsy (EUS-FNB) or metastatic biopsy of patients with pancreatic ductal Adenocarcinoma (PDAC).PDAC is the most common type of pancreatic cancer (>90%) and has an extremely high mortality rate.PDAC organoid retains the complex morphological characteristics of the primary tumor (different degrees of heterogeneous hyperplasia columnar cells, similar to low-level PanIN), genomic characteristics (KRAS>90%, TP53 50-75%, SMAD4 55%, CDKN2A 25% mutation rate) and histological characteristics.PDAC organoid exhibits a dense and irregular budding structure in morphology, which contrasts with the cystic structure of normal pancreatic duct organoid.
