Human Lung Organoids (Human Lung Organoids) are 3D self-organizing structures established from human lung tissue, which can be derived from surgically removed lung tissue (remote pulmonary parenchyma area), bronchial-alveolar lavage fluid, or fetal lung tissue.Pulmonary organoids contain proximal airway cells (basal cells, ciliated cells, cup-shaped cells, club cells) and distal alveolar cells (AT2 cells and AT1 cells), reflecting the complex structure and cellular diversity of the lungs.In 2013, Barkauskas et al. first reported that AEC2 cells are the main stem cells of the alveoli, and co-culture with PDGFRa+mesenchymal cells can form alveolospheres (alveolospheres) containing AEC2 and AEC1.Nikolić et al. (2017) established a long-term self-renewing organoid from the tip of the human fetal lung bud, retaining the dual expression of SOX2/SO9.Human pulmonary organs can be used to simulate diseases such as lung development, pulmonary fibrosis, COPD and COVID-19.
