Colorectal cancer (CRC) is the third most common malignant tumor in the world.The development of CRC follows the "adenoma-adenocarcinoma sequence", which involves progressive mutations in key driver genes such as APC, KRAS, SMAD4, and TP53.According to molecular characteristics, it can be divided into four consensus molecular subtypes: CMS1 (microsatellite unstable/immune type), CMS2 (typical/WNT-driven type), CMS3 (metabolic type) and CMS4 (interstitial type).Patient-derived CRC organoids can accurately reproduce the histopathological characteristics, gene mutation profiles and drug response of the primary tumor, and are one of the most important preclinical models in CRC research.Compared with normal intestinal organoids, CRC organoids usually exhibit constitutive activation of the Wnt pathway (such as APC mutations or β-catenin activation mutations), so they can grow without exogenous Wnt3a and R-spondin.
